543 Chronic wound environment shapes virulence of human commensal bacteria

چکیده
Staphylococcus epidermidis is one of the most abundant skin-commensal known to modulate cutaneous immune response. Emerging evidence suggests S. isolates from healthy skin improve barrier integrity and response wounding. However, could also carry a reservoir antimicrobial resistance genes(ARGs), adding this microbe list “accidental” pathogens. Hence, we aimed characterize chronic wounds (CW) evaluate their virulence potential effect on wound healing. Shotgun metagenomic sequencing was performed analyze presence ARG genes in both environments. Furthermore, susceptibility tested using microdilution method. To assess traits selected isolates, biofilm formation adhesion components extracellular matrix (ECM) were performed. Human ex vivo model used Results pointed prevalence CW associated with gentamicin, ampicillin, erythromycin, norfloxacin, tetracycline, trimethoprim resistance. This functionally confirmed, showed higher minimal inhibitory concentration (MIC) values for these antibiotics benzalkonium chloride, widely disinfectant. All strains exhibited ability bind ECM compared strains. feature correlates high vitro assays. Infection human increased accumulation bed suggesting strong formation. Our study that microenvironment influenced selection capacity form biofilm. data reflects dangers antibiotic overuse due frequency virulent found CW.

Ciprofloxacin dosage and emergence of resistance in human commensal bacteria.
BACKGROUND Although optimization of the fluoroquinolone dosage increases the efficacy of this class of drugs against bacterial infections, its impact on the emergence of resistance in commensal bacteria is unknown. METHODS Six different 14-day dosages of oral ciprofloxacin were randomly assigned to 48 healthy volunteers. Individual pharmacokinetic and pharmacodynamic parameters combining anti...

متن کاملCommensal ocular bacteria degrade mucins.
BACKGROUND/AIMS Antimicrobial activity in tears prevents infection while maintaining a commensal bacterial population. The relation between mucin and commensal bacteria was assessed to determine whether commensals possess mucinolytic activity, how degradation depends on mucin integrity, and whether mucins affect bacterial replication. METHODS Bacteria were sampled from healthy eyes and contac...
متن کاملClinical identification of bacteria in human chronic wound infections: culturing vs. 16S ribosomal DNA sequencing
BACKGROUND Chronic wounds affect millions of people and cost billions of dollars in the United States each year. These wounds harbor polymicrobial biofilm communities, which can be difficult to elucidate using culturing methods. Clinical molecular microbiological methods are increasingly being employed to investigate the microbiota of chronic infections, including wounds, as part of standard pa...
